Properties: Non-hemolytic, enzyme-compatible, low-foaming, and acts as an effective wetting agent, with additional emulsifying, stabilizing, and dispersing functions. It is readily soluble in water-alcohol mixtures and aqueous-organic solvent blends. Stable in acidic and neutral solutions but decomposes in alkaline conditions. Its surface-modifying properties help reduce surface tension while enhancing absorbency and permeability. AEROSOL OT also exhibits partial antistatic and softening effects.

Mechanism of Action

TMB Stability Control:

Terminates Oxidation Reaction: Aerosol OT forms a stable dark blue complex with TMB (3,3',5,5'-tetramethylbenzidine), preventing its further oxidation to the brown end product (quinoneimine form).

Enhances Oxidized State Stability: The complex remains in the highest oxidation state (TMB⁺⁺), significantly prolonging the chromogenic duration of TMB and preventing signal decay.

Improved Solubility and Compatibility:

Enables the TMB-TMB complex to be compatible with both aqueous and organic solvents (e.g., toluene), broadening the applicability of detection methods (e.g., organic solvent extraction or biphasic reaction systems).

ELISA (Enzyme-Linked Immunosorbent Assay): Stabilizes the TMB substrate, ensuring consistent and prolonged color development.

Chemiluminescence Kits: Enhances luminescence efficiency and signal stability.

POCT (Point-of-Care Testing) Devices: Maintains TMB activity in microplates or test strips, reducing inter-batch variability.

Reference Dosage

ELISA Chromogenic Solution: 0.1%-0.3% (added after stopping the reaction)

Chemiluminescence Substrate: 0.05%-0.2% (pre-mixed into the luminescence system)

Microplate Pretreatment: 0.01%-0.05% (for surface wetting before coating)

Application Scope

TMB-HCl chromogenic solutions (e.g., ELISA stop solution), chemiluminescence substrate systems, microplate pretreatment solutions.

Biotin/avidin labeling systems: Enhances conjugation efficiency.

Precautions

Storage Conditions: Store sealed and protected from light. Avoid moisture and ethanol evaporation, which may cause gelation (if gelled, add 1%-2% ethanol to redissolve).

Recommended Container Materials: Stainless steel, aluminum, or glass-lined containers (avoid long-term contact with plastic).

pH Range: Use only under acidic/neutral conditions (pH ≤ 7). Decomposes and becomes ineffective in strongly alkaline environments.

Temperature Adaptability: Freeze-thaw cycles do not affect activity, but ensure thorough mixing before use.

Avoid direct mixing with strong oxidizing agents (e.g., high-concentration H₂O₂), as this may cause complex dissociation.

When used in protein-containing systems (e.g., serum samples), validate whether it affects antibody binding activity.
