Fluoroquinolones are a class of synthetic antibacterial agents belonging to the third and subsequent generations of quinolone drugs. They exert their effect by inhibiting bacterial DNA gyrase, thereby interfering with DNA replication. This class demonstrates broad-spectrum antibacterial activity, particularly against Gram-negative bacilli and some Gram-positive cocci. The introduction of a fluorine atom in 1979 with norfloxacin (third-generation) enhanced lipid solubility and antibacterial efficacy. Subsequently developed agents like ciprofloxacin and ofloxacin further optimized the antimicrobial spectrum and pharmacokinetic properties.These drugs exhibit good oral absorption and excellent tissue penetration, making them suitable for treating urinary tract, respiratory, and soft tissue infections.
