Soybean 11S Globulin, also known as Glycinin (officially designated as Gly m 6), is the second core allergen in soybeans and a major seed storage protein. It constitutes approximately 40% of the total soybean seed protein and over 70% of the total allergenic protein content. With a molecular weight ranging from 300 to 380 kDa, it is a storage protein characterized by exceptional thermal stability. Even after conventional cooking and processing, it retains its allergenicity and exhibits greater resistance to processing compared to the 7S globulin.

It primarily triggers allergies via IgE-mediated Type I hypersensitivity reactions. Upon ingestion, the immune system identifies the protein as a foreign invader, stimulating the production of specific IgE antibodies that bind to mast cells and basophils. Subsequent exposure leads to rapid IgE cross-linking, triggering cellular degranulation and the release of inflammatory mediators like histamine, resulting in allergic symptoms. In livestock farming, it can also induce Type III hypersensitivity reactions, causing intestinal immune damage, and is classified as a typical antinutritional factor.

Glycinin is one of the most scrutinized allergens in pediatric soy allergies and plant-based food processing. Due to structural similarities, individuals allergic to peanut 11S globulin may also experience cross-reactivity with soybean 11S globulin.
