Staphylococcal Enterotoxin B (SEB) is a heat-stable exotoxin produced by Staphylococcus aureus. As a member of the B serotype, it is a critical causative agent of staphylococcal food poisoning. Additionally, SEB serves as a standard model toxin in biological research and has historically been investigated as a potential biowarfare agent.
SEB is secreted by pathogenic Staphylococcus aureusstrains harboring the sebgene and is a core member of the staphylococcal enterotoxin family (SEA–SEE). This soluble protein toxin (~28 kDa) exhibits the strongest thermostability among all serotypes; heating at 100°C for 30 minutes fails to achieve complete inactivation. Furthermore, SEB resists proteolytic hydrolysis in the gastrointestinal tract. Consequently, conventional cooking cannot fully eliminate its toxicity, and ingestion of preformed toxin—even in the absence of viable bacteria—leads to illness.
Transmission occurs primarily via consumption of contaminated foods. High-risk vectors include unrefrigerated dairy products, ready-to-eat meats, and cakes rich in starch or protein.
SEB is a classic superantigen. Unlike conventional antigens, it does not require processing by antigen-presenting cells but instead non-specifically activates massive T-cell proliferation. This dual capability allows it to both stimulate the proliferation of peripheral blood mononuclear cells and induce immunosuppression. Notably, recombinant SEB mutants (e.g., D9N) have demonstrated definitive inhibitory effects on transplanted tumors such as hepatocellular carcinoma and sarcoma, and are currently under investigation for antitumor applications.
