Staphylococcal Enterotoxin A (SEA) is a heat-stable protein toxin produced by Staphylococcus aureus. As a member of the A serotype, it is the most prevalent causative agent of staphylococcal food poisoning.

SEA is secreted by pathogenic S. aureusstrains harboring the seagene. Among the eight classic serotypes (A, B, C1, C2, C3, D, E, F), SEA accounts for the majority of food poisoning outbreaks. The toxin proliferates readily in nutrient-rich substrates high in protein or starch. Common high-risk contaminated foods include dairy products, ready-to-eat meats, leftover rice, and chilled processed cakes.

As a prototypic superantigen, SEA possesses a crystalline structure comprising two distinct domains: a β-barrel and a β-grasp motif. Its overall architecture shares high homology with Staphylococcal Enterotoxin B (SEB) and Toxic Shock Syndrome Toxin-1 (TSST-1).

SEA is a classic preformed toxin, meaning ingestion alone causes disease without requiring viable bacteria. Its pathogenesis involves two pathways:

Direct Neural Action: It binds to neural receptors in the enteric nervous system, stimulating the vomiting center to trigger severe gastrointestinal reactions.

Superantigenic Activity: Unlike conventional antigens, SEA bypasses antigen-processing by antigen-presenting cells. It directly binds to Major Histocompatibility Complex (MHC Class II) molecules, leading to the non-specific activation of a massive population of T lymphocytes and triggering a systemic immune response.
