Specific interference is primarily caused by heterophilic antibodies (human anti-animal antibodies), which can lead to significant false-positive or false-negative results. It is reported that such interference can occur in 30-40% of patient samples. These heterophilic antibodies may be IgAs, IgEs, IgGs, IgMs, or Ig(n). They can exist as polymers or monomers and vary widely in concentration. However, they all specifically target the antibodies used in the detection system.
Since most immunoassays utilize monoclonal mouse antibodies, human anti-mouse antibodies (HAMA) are the main cause of detection interference.
Interference elimination is a critical task in the development of immunoassay kits. Achieving higher sensitivity, greater precision, and more convenient operation in detection systems requires effective interference elimination.
The design of the assay significantly influences interference elimination. For example, IgG1 detection requires the elimination of IgG1 interference, just as IgG2a detection requires the elimination of IgG2a interference. HBR2 is an IgG1 monomer blocker.
HBR2 can eliminate HAMA, rheumatoid factor (RF), and other heterophilic interferences that may cause false-positive or false-negative results.
