Estradiol Benzoate is an estrogenic drug. It is metabolized in the liver to estradiol. After binding to sex hormone-binding globulin (SHBG), the unbound free fraction acts on target tissues, producing estrogenic effects. These include promoting endometrial proliferation, enhancing uterine smooth muscle contraction, and stimulating breast development and hyperplasia. At high doses, it can inhibit prolactin release, counteract androgenic effects, and increase calcium deposition in bone. It is clinically used to treat estrogen deficiency conditions such as atrophic vaginitis, menopausal syndrome, and ovarian insufficiency.
