In the normal brain, the cortex and hippocampus strongly express APP and Aβ. The primary source of Aβ is neurons, predominantly released from synapses. Endogenous Aβ mainly originates from excitatory neurons. Within neurons, Aβ is located in dendrites and axon terminals, while at synapses, it is primarily distributed in the presynaptic membrane.In healthy individuals, Aβ is a normal, soluble neuronal metabolite that regulates several key physiological functions and acts as a neuromodulator for synaptic plasticity, memory, and neurotransmitter release. The transition from physiological to pathological states is associated with an abnormal increase in Aβ levels, resulting from an imbalance between Aβ production and clearance.
